Nicotinamide adenine dinucleotide, commonly abbreviated as NAD+, is a naturally occurring coenzyme present in all living cells. It is composed of two nucleotides joined through their phosphate groups, with one nucleotide containing adenine and the other containing nicotinamide, a derivative of vitamin B3.
NAD+ plays a central role in cellular energy research because it participates in oxidation-reduction reactions that allow cells to transfer energy from nutrients into biochemical processes. During these reactions, NAD+ accepts electrons and is converted into its reduced form, NADH. NADH can then donate those electrons during additional metabolic reactions before being converted back into NAD+.
Because of this reversible NAD+/NADH cycle, NAD+ is widely studied in connection with mitochondrial function, cellular respiration, ATP production, glucose metabolism, fatty-acid metabolism, and overall cellular energy regulation.
NAD+ also serves as a required substrate for several classes of enzymes involved in cellular maintenance and signaling. These include sirtuins, poly-ADP-ribose polymerases, commonly called PARPs, and other NAD-dependent enzymes studied in relation to DNA repair, gene expression, oxidative stress, mitochondrial biology, cellular aging, and metabolic regulation.
NAD+ has the molecular formula C21H27N7O14P2, a molecular weight of approximately 663.43 g/mol, and CAS number 53-84-9. It exists primarily in two interconvertible forms: the oxidized form, NAD+, and the reduced form, NADH.
